Classification of PCR machine products

According to the purpose of DNA amplification and the criteria of detection, PCR machines can be divided into three categories: general PCR machines, gradient PCR machines, and in situ PCR machines.

1. Ordinary PCR machines

PCR machines that can only run one specific annealing temperature for one PCR amplification are called conventional PCR machines, also called ordinary PCR machines. Multiple runs are required if different annealing temperatures are to be done. It is mainly used for simple, annealing temperature amplification of target genes. This instrument is mainly used in scientific research, teaching, medical clinical, inspection, and quarantine institutions.

2. Gradient PCR machine

A PCR machine that can set a series of different annealing temperature conditions (temperature gradients, usually 12 temperature gradients) for single PCR amplification is called a gradient PCR machine. Because different DNA fragments being amplified have different optimal annealing temperatures, by setting a series of gradient annealing temperatures for amplification and thus one-time PCR amplification, the optimal annealing temperature with high expression can be selected for effective amplification. PCR machines are also used to study the amplification of unknown DNA at annealing temperature, which saves cost and time at the same time. The gradient PCR machine can also do normal PCR amplification without setting the gradient. It is mainly used in research and teaching institutions.

3. In situ PCR machine

The intracellular gene amplifiers used for localization analysis from intracellular target DNA are called in situ PCR machines. Such as the location of the pathogen gene in the cell or the location of the target gene's action in the cell. It is to maintain the integrity of cells or tissues so that the PCR machine reaction system penetrates into tissues and cells and performs gene amplification at the location of target DNA in cells. PCR machines not only detect target DNA but also mark the location of target sequences within cells, which is of great practical value for studying the pathogenesis of diseases and the transformation of clinical processes and pathologies at the molecular and cellular levels. It is mainly used in clinical and scientific research.

Clinical applications of PCR machines

1. Diagnosis of infectious diseases: PCR machines are particularly suitable for the detection of pathogens with long culture cycles or lack of stable and reliable detection methods in infectious diseases.

2. Diagnosis of hereditary diseases: the pathogenesis of hereditary diseases is based on the structural variation of nucleic acid molecules and expression products of nucleic acids, such as protein or enzyme molecular structure changes. the principle of PCR machine provides an effective means to detect this type of disease.

3. Diagnosis of malignant tumors: PCR machines are used for the detection of oncogenes and oncogenes deletions and point mutations as well as the detection of tumor-related viral genes, bringing a simple, rapid, and accurate method for clinical diagnosis, while PCR machines also provide a means to monitor the treatment and prognosis of tumor-related diseases.

4. Transplant typing: With the emergence of PCR machines, molecular biology technology has been introduced into the field of HLA typing. Through PCR machine amplifiers, a rapid and accurate HLA genotyping method can be established to meet the needs of clinical transplant typing.

The working environment of the PCR machine

Temperature: 5℃~40℃; relative humidity: not more than 80%; clean environment with little dust and avoid direct sunlight; PCR machine should be far from the heat source, water source, and strong electromagnetic interference source.
